Sci-fi thriller "All You Need Is Kill" - also starring Emily Blunt - is being made at the £100 million pound site.
Chancellor George Osborne has confirmed the news.
He says many of the jobs will be in digital and special effects.
The studio complex is no a stranger to blockbusters - James Bond, Star Wars and Harry Potter have all been filmed there.
Cruise has already started pre-production on the film, which is based on
a Japanese novel by Hiroshi Sakurazaka, after finishing work in Iceland
on Oblivion.
The actor will star as a futuristic soldier who becomes caught in a time
loop of his last day in battle as he fights a group of invading aliens.
